Prospect of electrophoresis technology

Compared with foreign countries, the clinical application of electrophoresis technology in China has a certain gap. According to the standards of foreign laboratories, domestic Class III A and Class III B hospitals need to be equipped with automatic electrophoresis devices, but many domestic Class III A hospitals still do not use such equipment. Electrophoresis projects generally only focus on serum proteins in China, while foreign projects include serum proteins, urine proteins, CK, LD, ALP, GGT isozymes, cholesterol / triglycerides of various lipoproteins, etc. For electrophoresis reagents used for analysis, agarose gel electrophoresis plates accounted for more than 95% in Europe and the United States in recent years, and 40% of the domestic market still uses cellulose acetate film electrophoresis plates, which is also a big gap from foreign countries. However, in recent years, some scientific electrophoresis technologies used in colleges and scientific research institutes, such as high-efficiency capillary electrophoresis, capillary electrophoresis chips, and electrophoresis-chromatography-mass spectrometry, have begun to be used in clinical laboratories. If a company manufacturer can develop special reagents with stable quality and clear related parameters like reagents on a biochemical analyzer, it will speed up its promotion and application process.

Judging from the requirements of current clinical laboratories, electrophoresis instruments must have the following functions:

(1) Fully automated: the machine must be able to work off the machine, and the machine can be completed from sampling to report.

(2) Fast speed: All projects should be completed within 60 minutes and also have emergency department projects such as CK, LD equivalent enzymes and subtype projects. Provide timely and accurate diagnostic information for the clinic.

(3) Complete project: In addition to traditional projects such as serum protein, lipoprotein, hemoglobin, CK, LD isozymes, it must also be able to complete urine protein, cerebrospinal fluid protein, immunofixation (monoclonal antibody), alkaline phosphatase (ALP) And (g-glutamyl transpeptidase (GGT) isozyme, cholesterol (high density / very low density / low density lipoprotein), lipoprotein (a) and other latest inspection items.

(4) High sensitivity: high sensitivity electrophoresis tablets must be used. For example, agarose gel system can obtain electrophoresis results with high accuracy and high resolution.

Newer studies have now reported that protein electrophoresis is used for the analysis of tear, saliva, and hair proteins. Moreover, the clinical progress in electrophoresis technology is also changing rapidly. In the mid-1990s, the sequencer was significantly improved, and clustered capillary electrophoresis replaced gel electrophoresis. The completion of the human genome framework in 2001 was completed in advance thanks to the emergence of multi-channel, clustered capillary gel electrophoresis technology. The recently developed capillary electrophoresis is a new type of zone electrophoresis, also known as capillary zone electrophoresis. It can detect urine samples, plasma, serum, cerebrospinal fluid, red blood cells, other body fluids or tissues, and living animal samples. It can separate proteins, peptides, amino acids, sugars, enzymes, DNA, oligonucleotides, viruses, small and biologically active molecules, ions, drugs and their metabolites. Widely used in clinical disease diagnosis, clinical protein analysis, clinical drug analysis, metabolic research, pathological research, isozyme analysis, PCR product analysis, DNA fragment and sequence analysis, etc. Because of its high efficiency, fast, sensitive, wide application range, few samples and automation, it is gradually accepted by the majority of clinical laboratories. With the rapid development of international and domestic science and technology and the rapid development of laboratory medicine, electrophoresis technology is also constantly developing and updating. I believe that with the continuous development of electrophoresis technology, it will have broad application prospects in clinical medicine and molecular biology.
